ABSTRACT

Abstract The aim of this study was to develop a kefir apple-based vinegar and evaluate this fermentation process using new methodology with Biospeckle Laser. Brazilian kefir grains were inoculated in apple must for vinegar production. In this study, the microbial community present in kefir, and correspondent vinegar, was investigated using Matrix Assisted Laser Desorption/Ionization - Time of Flight Mass Spectrometry (MALDI-TOF MS) technique. Saccharomyces cerevisiae, Lactobacillus paracasei, Lactobacillus plantarum, Acetobacter pasteurianus and Acetobacter syzygii were the microbial species identified. S. cerevisiae, L. plantarum, A. pasteurianus and A. syzygii were found in smaller quantities at the beginning of the alcoholic fermentation, but were found throughout the alcoholic and acetic fermentation. Kefir grains were able to utilize apple must as substrate to produce ethanol, and acetic acid. Acetate, volatile alcohols and aldehydes in the vinegar-based kefir were also produced. The yield of acetic acid in the kefir vinegars was ∼79%. The acetic acid concentration was ∼41 g L-1, reaching the required standard for the Brazilian legislation accepts it as vinegar (4.0% acetic acid). Kefir vinegar showed good acceptance in the sensory analysis. The technology proposed here is novel by the application of immobilized-cell biomass (kefir grains) providing a mixed inocula and eliminating the use of centrifuge at the end of the fermentative process. This step will save energy demand and investment. This is the first study to produce apple vinegar using kefir grains.

ABSTRACT

Abstract Pediococcus acidilactici strain S1, a lactic acid-fermenting bacterium, was isolated from makgeolli-a Korean traditional fermented alcoholic beverage. Here we report the 1,980,172 bp (G + C content, 42%) genome sequence of Pediococcus acidilactici strain S1 with 1,525 protein-coding sequences (CDS), of which 47% could be assigned to recognized functional genes. The genome sequence of the strain S1 might provide insights into the genetic basis of the lactic acid bacterium with alcohol-tolerant.

ABSTRACT

A partir de cuatro muestras de pulque (bebida mexicana prehispánica) en diferentes etapas de fermentación denominadas aguamiel, semilla, contrapunta y corrida; se aislaron e identificaron un hongo levaduriforme, un cocobacilo Gram negativo y un bacilo Gram positivo pertenecientes a los géneros Saccharomyces sp, Zymomonas sp, y Lactobacillus sp. El consorcio desarrollado fue el responsable de la fermentación, alcohólica, ácida y viscosa propias de esta bebida tradicional Mexicana. El contenido alcohólico varió en función del tiempo de fermentación alcanzando porcentajes de 10.35 (v/v) y 9.01 (v/v) determinados por cromatografía de gases y espectroscopia Raman para la etapa denominada ®corrida¼. El producto terminado tiene un elevado contenido de proteínas (1 g/L), azúcares reductores (4.75 g/L) y poblaciones microbianas en ordenes de 45x108 UFC/mL para Saccharomyces sp, 41x107 UFC/mL para Zymomonas sp y 34x106 UFC/mL para Lactobacillus sp, que le dan a la bebida un elevado valor nutricional y los microorganismos aislados podrían tener un efecto benéfico sobre sistema digestivo al ser consumido por vía oral.

ABSTRACT

Embalagens práticas e visualmente atrativas estimulam o consumo de alimentos fora do lar. O risco à saúde do consumidor, decorrente do contato direto dessas embalagens com a boca, não tem sido avaliado. Foram examinadas latas de cerveja e de refrigerante (n=120), adquiridas no comércio na cidade do Rio de Janeiro. Elevados níveis de microrganismos mesófilos aeróbios, fungos, coliformes, enterococos e estafilococos coagulase-positiva foram encontrados, sendo freqüentemente maiores nas embalagens adquiridas de vendedores ambulantes. A presença de Salmonella sp., em duas embalagens provenientes de cada tipo de comércio, acenou como fator de elevado risco de infecção no trato intestinal. Processos de limpeza das latas, freqüentemente utilizados pelos consumidores, não revelaram eficiência na remoção dos microrganismos.

Desenvolveram-se dois meios de cultura para quantificação das bactérias microaerofílicas deteriorantes obrigatórias da cerveja (bmdoc), denominados de Odebrecht (O) e Odebrecht + Vitaminas (O + V), com o objetivo de preencherem os requisitos de crescimento rápido e conseqüente redução do período de incubação, crescimento de todo o espectro de cepas de bmdoc e inibição das demais bactérias presentes na cervejaria. Selecionaram-se 13 cepas de bmdoc, que foram inoculdas em cerveja comercial do tipo Pilsen, incubadas por 60 dias a 27ºC, durante três gerações.
